Somewhat newbie question
I wouldn’t start at 50. Try 5 rucks @20lbs 5@25 5@30 Etc. This will let you get your kit dialed in and your body used to the frequency. The Osprey will work. Just like backpacking the advice is to get the weight high and close to your back. This will take some fiddling with a large pack like yours.

Padded Hip Belt for Rucker 4.0.

How can I prevent my backpack unzipping itself?
Buy a quality ruck with a waist belt especially if you are going to be exercising with it. For rucking plus working out I use a GORUCK Rucker 4.0. The belt is sold separately so is a plate that fits the holder. You can get a plate on Amazon for a lot cheaper but make sure it’s compatible.

Advice on weighted vests
Backpack is better for rucking/distance. Vest is good for calisthenics or people who need to train to move in vests (military, some law enforcement). I have a GoRuck Rucker 4.0 and a cheap Amazon plate carrier with 20 and 40 lb plate pairs. I find the vest really compresses my spine. Backpack helps
